چکیده

Suspension cultures of Picea abies (L.) Karst released polymeric material into the culture medium when treated with an elicitor preparation from the spruce needle pathogen Rhizosphaera kalkhoffii. The presence of lignin (about 35%, w/w) was demonstrated by phloroglucinol/HCI reactivity and quantitation with thioglycolic acid. Carbohydrate (about 14%, w/w) and protein (about 32%, w/w) were also detected. Amino acid analysis revealed that hydroxyproline and proline predominated. Thioacidolysis and subsequent Raney nickel desulfurization allowed the analysis of lignin-building units and interunit bonds. Compared with spruce wood lignin, an approximately 20-fold higher relative amount of p-hydroxyphenyl units was determined. A high content of p-hydroxyphenyl units is typical for certain developmental lignins, such as conifer compression wood and middle lamella lignins, as well as all induced cell culture lignins so far analyzed. Cross-linkages of the pinoresinol type ([beta]-[beta]) in the excreted cell culture lignin were markedly increased, whereas [beta]-1 interunit linkages were decreased relative to spruce wood lignin. The amount and nature of cross-linkages were shown to be intermediate between those in wood lignin and in enzymatically prepared lignins. In summary, the elicitor-induced stress lignin was excreted as a lignin-extensin complex that closely resembled early developmental lignins.

Condensed lignins are synthesized in poplar leaves exposed to ozone.

Poplar (Populus tremula x alba) trees (clone INRA 717-1-B4) were cultivated for 1 month in phytotronic chambers with two different levels of ozone (60 and 120 nL L(-1)). Foliar activities of shikimate dehydrogenase (EC 1.1.1.25), phenylalanine ammonia lyase (EC 4.3.1.5), and cinnamyl alcohol dehydrogenase (CAD, EC 1.1.1.195) were compared with control levels. In addition, we examined lignin con...

Polymerization of Different Lignins by Laccase

In this study the oxidative polymerization of different lignins, i.e. Flax Soda lignin, Spruce EMAL, and Eucalyptus Dioxane lignin by Trametes hirsuta laccase was compared. Initially the structures of the different lignins were compared by Fourier transform infrared spectroscopy. The reactivity of laccase with the different types of lignins in the absence of mediators was examined and verified ...

Quantitative iTRAQ proteome and comparative transcriptome analysis of elicitor-induced Norway spruce (Picea abies) cells reveals elements of calcium signaling in the early conifer defense response.

Long-lived conifer trees depend on both constitutive and induced defenses for resistance against a myriad of potential pathogens and herbivores. In species of spruce (Picea spp.), several of the late events of pathogen-, insect-, or elicitor-induced defense responses have previously been characterized at the anatomical, biochemical, transcriptome, and proteome levels in stems and needles. Howev...

Concomitant changes in viscoelastic properties and amorphous polymers during the hydrothermal treatment of hardwood and softwood.

The aim of this study was to understand how the molecular structures of amorphous polymers influence wood viscoelastic properties. Wood from oak and spruce was subjected to hydrothermal treatments at 110 or 135 degrees C. Wood rigidity, reflected by the wood storage modulus, showed different modification patterns according to the wood species or the temperature level. Because viscoelasticity is...

Activation of Lignin Biosynthetic Enzymes During Internodal Development of Aeluropus littoralis Exposed to NaCl

Lignin is one of the major characteristics of plant secondary cell wall that provides structural rigidity for the cells and tissues and hydrophobicity to tracheary elements. Internode tissues of Aeluropus littoralis as a halophyte grass were sampled at different developmental stages (from the first to the fifth internodes ) and under different NaCl concentrations. The influences of NaCl and int...
